Improved automated determination of bound N-acetylneuraminic acid in serum.

چکیده
The method in which thiobarbituric acid is used for sialic acid determination was adapted to a fully automated procedure, in which the chromogen need not be extracted with an organic solvent. The entire procedure, including hydrolysis, requires 35 mm and 25 l of serum for one analysis, but 30 samples can be analyzed per hour. The procedure, slightly modified, can be used to measure as little as 10 mg of bound sialic acid per liter.

Automated method for determination of bound N-acetylneuraminic acid in serum.
The usefulness of determining protein-bound sialic acid in serum is obvious, because a significant increase in various serum sialoglycoprotein concentrations (haptoglobin, orosomucoid, a1-antitrypsin, and ceruloplasmin) has been observed in many pathological states (1, 2). We earlier described(3) a fully automated method in which thiobarbitunc acid is used as a reagent. This very sensitive method...

متن کاملNew enzymatic determination of sialic acid in serum.
This enzymatic method for determination of sialic acid involves use of neuraminidase (EC 3.2.1.18), N-acetylneuraminate lyase (EC 4.1.3.3), acylglucosamine 2-epimerase (EC 5.1.3.8), N-acetylhexosamine oxidase (from Pseudomonas sp.), and peroxidase (EC 1.11.1.7). Because the method does not require pyruvic acid in the assay medium, interference by pyruvic acid in serum can be avoided. This simpl...

Objective(s) Some properties of neuraminidase produced by Pseudomonas aeruginosa PAO1 growth in a defined medium (BHI) were examined and evaluated for its features. Materials and Methods The obtained supernatant enzyme of P. aeruginosa PAO1 cultures was used in a sensitive fluorometric assay by using 2'-(4-methylumbelliferyl) a-D-N acetylneuraminic acid as substrate. As hydrolyzing MUN with ...
متن کاملRelation between raised concentrations of fucose, sialic acid, and acute phase proteins in serum from patients with cancer: choosing suitable serum glycoprotein markers.
Serum concentrations of fucose, sialic acid, and eight acute phase proteins were measured in single specimens from patients with cancer in order to determine whether the raised concentrations of protein bound sugars commonly found in cancer correlate with increased concentrations of the acute phase proteins. Strong positive correlations were found only with alpha 1-acid glycoprotein, alpha 1-an...
